PRODUCTION OF RAT LIVER CYTOCHROME P-450C

摘要

PURPOSE:To efficiently obtain the subject oxidase usable for various kinds of oxidation reactions, having one atom oxygen addition activity by culturing transformant yeast having a gene encoding rat liver cytochrome P-450c having a specific amino acid sequence. CONSTITUTION:Recombinant plasmid pAU157 having a gene encoding rat liver cytochrome P-450c having an amino acid sequence of the formula is treated with a restriction enzyme to isolate P-450c gene, which is integrated into manifestation vector pAAH5 having yeast alcohol dehydrogenase promoter and terminator to constitute recombinant plasmid pAMC1 containing a gene encoding rat liver cytochrome P-450c. The recombinant plasmid pAMC1 is transduced into yeast, which is transformed into transformant yeast Saccharomyces cerevisiae AH22 strain (FERM P-7,752). Then the yeast is cultured to efficiently give the objective rat liver cytochrome P-450c having one atom oxygen addition activity.
