PURPOSE:To industrially and advantageously obtain the subject substance useful as a drug, etc., for treating and preventing cardiopathy by treating urine with an adsorbent, such as polystyrene resin, fractionating the resultant eluate with a resin in the reverse phase, passing the obtained fraction through a gel filtration resin and separating a substance having digitaris-like action. CONSTITUTION:Human urine is treated with an adsorbent containing polystyrene or acrylic resin and methanol is passed therethrough to elute adsorbed substances. The methanol is distilled away under reduced pressure and the resultant residue is dissolved in water, then passed through a column of a resin in the reverse phase and adsorbed thereon. The adsorbed substances are subsequently eluted with a 20% aqueous solution of acetonitrile and the acetonitrile is distilled away. Fractionation is then carried out using reversed phase high performance liquid chromatography. The resultant active fraction is passed through a gel filtration resin and fractionated to afford the objective novel physiologically active substance capable of exhibiting <3>H-ouabain-binding inhibitory activity against digitalis receptors on erythrocytic surfaces. |
