PURPOSE:To make possible to obtain a kestose in high yield by culturing bioorganism of genus Scopulariopsis(SCP) digesting glucose in medium containing sucrose. CONSTITUTION:The SCP SP HS-0002 strain (A) is collected by isolating from soil of SHARI-CHO, SHARI-GUN in HOKKAIDO (name of a district). Said strain has mycological properties such as 10-30mum length and 2.5-3.0mum breadth of conidiophore and 20-30 deg.C optimum growing temperature, etc., produces kestose in the presence of sucrose and digests glucose. Next, said strain (A) is cultured in a medium containing 5-50wt.% sucrose, 1-4wt.% peptone, etc., and 0.1-1.0wt.% MgSO4, etc., at pH5-8 and 24-40 deg.C for 72-90 hour with ventilation and agitation to obtain a cultured substance (B). Then, the component (B) is filtered and the strain is separated, thus resultant filtered solution is heated at 60-80 deg.C and reducing sugar, etc., is decomposed by adding CaO, etc., then the solution is filtered, thus resultant filtered solution is purified by treatment of ion-exchange resin or active carbon, etc., to recover kestose in about 80% yield. |
