RECOMBINANT PLASMID FOR BACTERIUM BELONGING TO GENUS THIOBACILLUS, MERCURY-RESISTANT VECTOR AND PREPARATION THEREOF

PURPOSE:To obtain a recombinant plasmid required to carry out gene manipulation in a bacterium belonging to the genus Thiobacillus containing a replicator of plasmid of bacterium belonging to the genus Thiobacillus and a replicator of vector plasmid of Escherichia coli. CONSTITUTION:A plasmid (e.g. pTSY91 or TSB121) containing a replicator of plasmid of bacterium belonging to the genus Thiobacillus is taken out from Thiobacillus ferrooxidans strain Y5-9 (FERM P-9157) and strain B-12 (FERM P-9156) and taken out. The plasmid and a vector plasmid of Escherichia coli (e.g., pBR322) are cleft by the same restriction enzyme, bonded and cloned in Escherichia coil to give a recombinant plasmid for a bacterium belonging to the genus Thiobacillus. A range containing a mercury-resistant gene site taken out from a plasmid (e.g. pME285) containing a mercury-resistant gene is integrated into a vector of Escherichia coli and the prepared recombinant vector plasmid is integrated into a fragment of the above-mentioned recombinant plasmid to give mercury-resistant shuttle vector plasmid.
