PURPOSE:To introduce a cyclic DNA into the cell of Acetobacter, in high efficiency, by carrying out the process in a solution containing bivalent metal. CONSTITUTION:A cyclic DNA is introduced into the cell of Acetobacter of the stage from the initial stage to the medium stage of the logarithmic period in a solution containing a bivalent metal selected from Ca, Mg, Sr, Ba, Mn and Zn and further containing a transformation-accelerating substance selected from polyethylene glycol, dimethylsulfoxide, N,N-dimethylformamide, protamine, polyornithine, polyvinyl alcohol, DEAE-dextran, sucrose and glycerol, and having a pH of 6.5-7.6. The cyclic DNA to be introduced is preferably a chimera plasmid obtained by introducing a drug-resistant plasmid to a plasmid separated from Acetobacter aceti No.1023. |
