PURPOSE:To prepare high purity acetic acid kinase in high efficiency, by adsorbing acetic acid kinase produced by the cultivation of acetic acid kinase-producing microorganisms with a water-insoluble carrier prepared by reacting a blue eye with a water-insoluble polymer. CONSTITUTION:Microorganisms capable of producing acetic acid kinase, e.g. Bacillus stearothermophilus, are inoculated and cultured in a conventional medium, and the treated microbinal cells are brought into contact with a water-insoluble carrier shown by formula I (one of R1 and R2 is H and the other is SO3Na group; R3 is water-insoluble polymer). The acetic acid kinase adsorbed to the carrier is eluted with e.g. a buffer solution containing 10mM of adenosine triphosphat, a buffer solution containing 10mM of adenosine triphosphate and 100mM of potassium chloride, etc. |
