Zur radioimmunologischen Bestimmung von Prokollagen (Typ III) und Prokollagen-Peptid (Typ III) werden eine bestimmte Menge radioaktiv markiertes Prokollagen (Typ III) oder Prokollagen-Peptid (Typ III) und ein hochspezifisches Anti-Prokollagen- (Typ III) -Serum bzw. Anti-Prokollagen-Peptid-(Typ III)-Serum gemeinsam mit einer Probe mit unbekanntem Gehalt an Prokollagen (Typ III) oder Prokollagen-Peptid (Typ III) zur Reaktion gebracht, der gebildete Antigen-Antikörper-Komplex wird abgetrennt, zweckmäßig durch Zusatz eines gegen das hochspezifische Antiserum gerichteten zweiten Antikörpers, und die Radioaktivität des Komplexes oder des Überstandes gemessen. Hierfür geeignetes hochgereinigtes Prokollagen-Peptid (Typ III) erhält man durch Abbau von Gewebe oder Körperflüssigkeiten mit Kollagenase und Reinigung durch Immunadsorption und Chromatographie.
