A method of detecting a Group A streptococcal analyte in a pathogenic streptococcal infection, comprising: a) providing a patient sample suspected of harboring Group A streptococci; b) contacting the sample with a PlyA polypeptide comprising an amino acid sequence with at least 85% sequence identity to SEQ ID NO: 2 and a purification tag, in an amount effective to lyse Group A streptococci if present in the sample; and c) detecting the presence or absence of a Group A streptococcal analyte present in the sample, wherein the presence of the Group A streptococcal analyte in the sample indicates the presence of a streptococcal infection in the subject.
