A method for manufacturing DNA polynucleotide which is isolated from semen or egg of fishes is provided to have DNA polynucleotide fragment with high yield by performing enzyme lysis process. A method for manufacturing a DNA polynucleotide fragment which is isolated from semen or egg of fishes comprises thawing process of the semen or egg, enzyme lysis process, sterilization process, molecular weight reduction process, precipitation process and dry-granulation process. The enzyme lysis process is performed at pH 7.0-7.4 and 43-47°C. The sterilization process is performed at 100-109°C for 10 to 30 minutes using weak acid. The molecular weight reduction process is performed at pH 4.0-4.4 and 68-72°C. The DNA polynucleotide fragment has molecular formula, C9.83H12.33N3.72O6.01PNa and 50-1500 kDa of molecular weight. The DNA fragment is hardly soluble in water and alkali, soluble in alcohol in alcohol and is insoluble in ether and acetone.
