PURPOSE: A method for enhancing long-term preservation efficiency of spermatogonial stem cells is provided to enhance the survival rate of the stem cells after thawing and to develop human infertility treatments. CONSTITUTION: A cryopreservation solution for spermatogonial stem cells contains 0.05-0.2M of trehalose. The solution is MEM-alpha containing 5-20% FBS and 5-20% DMSO. A method for cryopreservation of spermatogonial stem cells comprises: a step of treating the testis of mammals with enzyme to isolate single cell groups; a step of collecting spermatogonial stem cells from the single cell groups; a step of culturing the spermatogonial stem cells in vitro and collecting; and a step of performing cryopreservation of the solution. |
