Provided is an isolated nucleic acid comprising a heterologous first nucleotide sequence of interest, and further comprising at least two second nucleotide sequences (introns) heterologous to the first sequence, comprising two sets of splice elements, such that the second intron is different from the first intron. The first intron imparts a biological function in the absence of the activity of splice elements in the second intron. The two introns are in tandem, or separated by 25-200 nucleotides, and the second intron can reside in the promoter or in 3' UTR of the first nucleotide sequence. The expression constructs are useful in providing a method of controlling gene expression by using blockage of the second set of splice element, e.g. by blocking oligonucleotides. Further provided are use of the compositions for the manufacture of medicaments and for in vitro methods of identifying compounds that block the second set of splice elements.
