A DNA marker is provided to be used as a biological marker capable of increasing the meat amount as well as preventing the deterioration of the meat quality by an SNP portion in a 5'-promoter region of a Myogenin gene related to a pig muscle cell differentiation, thereby being useful for a pig breeding industry due to early selection and productivity improvement. A composition for detecting increase of pig muscle-cell number consists of a DNA fragment including a 788th T base in a sequence of SEQ ID : NO. 9. A primer for searching an SNP related to muscle-cell number of a 5'-promoter region of a Myogenin gene of a pig consists of oligonucleotides described as SEQ ID : NOs. 7 and 8. A kit for analyzing SNP related to the muscle cell number increase of a pig comprises the primer, a PCR reaction mixture and BspCNI, which is a restriction enzyme for analyzing RFLP. A method for identifying the muscle cell number increase of a pig comprises the steps of: (a) isolating a genomic DNA from the pig; (b) amplifying an SNP of a 5' promoter region of a Myogenin gene using the DNA as a template; (c) treating the amplified DNA with BspCNI; and (d) comparing RFLP(restriction fragment length polymorphism) of gene fragments obtained from the step (c).
