摘要 <p>#CMT# #/CMT# Osseous substitute presenting an activity on osseous regeneration, comprises fucan presenting a weight average molar mass of 5000-100000 g/mol, preferably 10000-40000 g/mol. #CMT# : #/CMT# INDEPENDENT CLAIM are included for: (1) the preparation of osseous substitute comprising covering the biomaterial surface by a coating of fucans; (2) a culture medium for a osteocompetent type cell of bone marrow, bone or periosteum, comprising fucans presenting a weight average molar mass of 5000-100000 g/mol, preferably 10000-40000 g/mol; (3) cell culture of osteocompetent cells of bone marrow, bone or periosteum, comprising cultivating the cells in the culture medium or in the osseous substitute; and (4) a medical device presenting an activity on osseous regeneration, comprising fucans presenting a weight average molar mass of 5000-100000 g/mol, preferably 10000-40000 g/mol. #CMT#USE : #/CMT# The fucans are useful in osseous grafting, osseous engineering and osseous regeneration. #CMT#ADVANTAGE : #/CMT# The fucans are able to promote and to accelerate, migration, proliferation of osteoblastic cells and the synthesis of one new osseous extracellular matrix and its mineralization. #CMT#BIOLOGY : #/CMT# Preferred Composition: The osseous substitute/culture medium/medical device further comprises growth factors such as fibroblast growth factors, transforming growth factors, insulin growth factors I, platelet derived growth factors, bone morphogenetic proteins or vascular endothelial growth factors. The osseous substitute further comprises cytokines comprising interleukin 1, interleukin 4, interleukin 6, TNF-alpha, granulocyte-macrophage colony-stimulating factor or macrophage colony-stimulating factor. The osseous substitute comprises osteocompetent cells from bone marrow, bone or periosteum. The medical device comprises the osseous substitute. Preferred Process: The biomaterial is impregnated with the fucans. The osseous substitute is colonized by osteocompetent cells from bone marrow, bone or periosteum. The osteocompetent cells of bone marrow, bone or periosteum are cultivated in a culture medium comprising fucans presenting a weight average molar mass of 5000-100000 g/mol, preferably 10000-40000 g/mol. Preferred Components: The fucans are obtained from brown algae. The osseous substitute comprises a biomaterial. The biomaterial comprises titanium, collagen, deproteinized and/or demineralized bone, coral, calcium phosphate ceramics, hydroxyapatite, beta tricalcium phosphate or bio-active glasses. The surface of the biomaterial presents a coating comprising the fucans. #CMT#EXAMPLE : #/CMT# Normal human osteoblasts were seeded in 24 culture box wells (20000 cells/well) containing the osseous biomaterials (bovine spongy osseous weft) optionally impregnated with fucans. The grafts were maintained in a mineralized culture medium containing Dulbecco's Modified Eagle's medium, fetal calf serum (10%), ascorbic acid (50 mu g/ml), insulin (5 mu g/ml) and transferrine (5 mu g/ml). The culture medium was renewed all the 3 days throughout the experiment. The grafts were obtained after 10 or 30 days of culture and withdrawn from culture box.</p>
