摘要 <p>In a homogeneous assay for the detection or quantification of an analyte, a chemiluminescent acridinium compound (A) is used. Independent claims are included for the following: (1) a homogeneous energy transfer sandwich assay for the detection or quantification of a macromolecular analyte (M1) involves: (i) conjugation of an antibody specific for (M1) with (A); (ii) conjugation of a second antibody specific for (M1) with an acceptor molecule (e.g. fluorophore or quencher); (iii) complexation of the conjugates with (M1) in a sample to from a reaction mixture; (iv) triggering the chemiluminescence of the binding complex reaction mixture by adding chemiluminescence triggering reagents in pH of 6 - 10 to transfer excited state energy from (A) of the first conjugate to the acceptor molecule of the second conjugate, causing light emission from the acceptor molecule or light attenuation; (v) measuring the amount of light with a luminometer; and (vi) calculating the concentration of the macromolecular analyte by comparing the amount of light emitted from the reaction mixture with a standard dose response curve which relates the amount of light emitted to a known concentration of the macromolecular analyte; and (2) inhibition of a pseudo-base of an acridinium ester formation involves attaching electron donating functional groups at C-2 and/or C-7 positions on the acridinium nucleus of an acridinium ester.</p>
