PROBLEM TO BE SOLVED: To massively obtain the subject compound capable of stimulating the differentiation of granulocytes and macrophages and effective for the therapy of tumors, etc., by culturing lymphocytes separated from leukocytes contained in the blood of a healthy person in a culture medium containing an inducing agent such as PHA and CMP. SOLUTION: This method for producing a factor for stimulating the colonies of granulocytes and macrophages (GM-CSF), effective for treating immunological function depression, tumors, etc., in a large amount comprises centrifuging blood collected from a healthy person to collect leukocytes, adding a phosphoric acid-buffered physiological saline solution (PBS) (specific gravity: 1.077) diluted into a four-fold volume to the collected leukocytes to separate the lymphocytes, washing the separated lymphocytes with a PBS having a pH of 7.4, further washing the lymphocytes with a culture solution, counting the cells, controlling the concentration of the cells to a concentration of 8×10<6> cells/ml with the culture solution, subjecting the cell suspension to a shaking culture treatment overnight, and further culturing the cultured product in a culture medium containing an inducing agent consisting mainly of one or more kinds of PHA (plant agglutinin: phytohemagglutinin), CMP(Calboxy methel pacyman), etc., for >=168hr. |
