PURPOSE:To obtain a new glucose oxidase and to provide a method for producing the same. CONSTITUTION:For the first time, an alkali-resistant glucose oxidase has been found in a cultured product of Coriolus hirsutus. The enzyme purified has high oxidation activity of glucose, and can also oxidize maltose, lactose, sorbose, mannose or xylose. This enzyme has the following characteristics: optimal pH value: 7, stable within the pH range of 4 to 12; optimal temperature: 35 deg.C; molecular weight: 68,000; and isoelectric point: 5.4+ or -0.2. The enzyme is inhibited by Cu<2+> or dithiothreitol, being strongly inhibited by esp. Hg<2+>, Mn<2+> or NaN3. There is no known enzyme having characteristics mentioned above; therefore, this enzyme is a new one. |
