The method comprises treating 5-thiazolinone deriv. (a) of amino acid with rhodamine deriv. amino cpd. H2N-X (b) to form phenylthiocarbamylamino acid deriv. (c), and detecting the aminoacid deriv. (c) with high sensitivity. In the formulae X= aminofluorescein or rhodamine deriv. amino cpd. R1= e.g. alkyl. Pref. detection of (a) is sensitively carried out by fluorophotometry. Amino acid deriv. can be simply detected with very high sensitivity without using radioactive isotope. The method is useful for deciding amino acid sequence from N-terminal of protein or peptide. The detecting sensitivity by the method is 10 fmol. (f=10power(-15)).
