PREPARATION OF LYMPHOTOXIN DERIVATIVE

PURPOSE:To prepare the subject compound useful as a cancer remedy in a large amount by not adding a protease-inhibiting agent when a fused protein of human lymphotoxin with any other protein is produced in a microorganism host and the fused protein is subsequently recovered. CONSTITUTION:A microorganism host is transformed with a manifestation plasmid containing a gene coding a fused protein prepared by fusing a site of human lymphotoxin on the N end side of the 19-lysine thereof with any other protein, followed by culturing the transformed host the prepare the fused protein. The manifested product is recovered from the cultured product without adding a protease-inhibiting agent, thereby cleaving the fused protein with a host cell-produced protease between the 19-lysine and 20-methionine of the human lymphotoxin to provide the objective compound free from the peptide on the N end side.
