PURPOSE: To obtain the subject conjugate capable of enhancing the antibody response of a host by converting a carboxyl group-containing capsular polysaccharide into a thiol derivative and subsequently binding the thiol derivative to a protein with a hetero-bifunctional cross-linking agent. CONSTITUTION: This polysaccharide-protein conjugate is obtained by treating a carboxyl group-containing capsular polysaccharide, such as the Vi capsular polysaccharide of Salmonella typhi, with cystamine or cysteamine and subsequently binding the formed thiol derivative to a protein with a hetero- bifunctional cross-linking agent, such as N-succinimidyl 3-(2-pyridylthio) propionate. The protein includes tetanus toxoid, diphtheria toxoid, cholera toxin, Hemophilus influenza b type protein and IgG. A composition containing the conjugate in a carrier is injected into a host (at least twice at two week intervals) to enhance the antibody response of the host.
