PURPOSE:To obtain a high purity HBe antigen from blood plasma positive to the HBe antigen with good yield, by using cation exchanger after using cephalose combined with HBe antibody and carrying out washing and elution by guanidine hydrochloride. CONSTITUTION:An aqueous solution containing HBe positive blood plasma or a HBe antigen is brought into contact with HBe antibody combined cephalose in 5-8pH and salt concentration of 0.001-0.3 ion intensity and the HBe antigen is adsorbed. Next, the HBe antibody combined cephalose adsorbed the HBe antigen is washed thoroughly by 0.5-1.5M guanidine hydrochloride solution and 2-5M guanidine hydrochloride solution is added and then, the HBe antigen is eluted. After obtained eluate is dialyzed by an aqueous solution having 5-9pH and low salt concentration of <=0.005M ion intensity, said solution is brought into contact with an anion exchange material brought to equilibrium by a buffer solution having 5-9pH and low salt concentration of <=0.05M ion intensity and the HBe antigen is obtained in unadsorption fraction. A detection reagent of the HBe antibody is obtained by sensitizing thus produced HBe antigen aqueous solution to a granular carrier. |
