1. A droplet actuator device for conducting droplet operations, comprising:
(a) a bottom substrate and a removable top substrate; (b) an array of droplet operations electrodes arranged on the bottom substrate; wherein the top substrate is a self-contained replaceable/removable cartridge for a droplet actuator-based assay and an interface material comprising a dielectric layer disposed between the bottom substrate and the cartridge in such a way as form a reverse impression of the array of droplet operations electrodes on the bottom substrate.
