PROBLEM TO BE SOLVED: To provide a method for determining and measuring specific ingredients in a biological specimen with a liquid reagent that is constituted of two or more reagents, produced by making an enzyme having dehydrogenation-oxidizing ability that acts on the biological ingredients, in the presence of an electron receptor, treating the produced reduced electron acceptor with an electron carrier and a reducing pigment to make formazan formed, and then determining the formazan, and makes both the "high degree of accuracy" and "stability" of the reagent compatible. SOLUTION: This method for determining and measuring the specific ingredient in the biological specimen uses tetrazolium or its compound as the reducing pigment, controlling the pH of the reagent that contains the reducing pigment to be 8.0 or lower, and controlling the pH of the reagent to be 7.5 or higher, when reacting.
