PROBLEM TO BE SOLVED: To provide a rapid and convenient method which enables use of a targeting vector comprising a large homologous region, capable of modifying an endogenous gene or a chromosomal gene locus in a eukaryotic cell.SOLUTION: The invention is a method for engineering and utilizing a large DNA vector to target via homologous recombination, and modify in an arbitrary desirable fashion, an endogenous gene and a chromosomal gene locus in a eukaryotic cell. The invention further provides a rapid and convenient method of detecting a eukaryotic cell in which a large targeting vector(LTVEC) for a eukaryotic cell is correctly targeted, and a desirable endogenous gene or chromosomal gene locus is modified. The invention further includes an organism containing the genetic modification, use of these cells to generate an organism itself, and a method of the use. |
