Improved preparation method of Decitabine

摘要

PURPOSE: A method for preparing decitabine with improved yield and purity is provided to minimize isomer formulation. CONSTITUTION: A method for preparing decitabine with improved yeild and purity comprises: a step of reacting 1,3,5-triacetyl-2-dioxyribose of chemical formula 2 with silyl group-induced 5-azacytosien in methylene chloride or EDC solvent under the presence of trimethylsilyl trifluoromethanesulfonate Lewis acid to prepare a compound of chemical formula 4;a step of preparing a compound using ethyl acetate to prepare a compound of chemical formula 5; and a step of preparing the decitabine from the compound of chemical formula 5, with deprotected decitabine.
