Cell-based screens for compounds that inhibit the accumulation of amyloids are described. For example, the herein described screens may be used to assay the formation, or inhibition, of pre-amyloid or amyloid aggregates and/or oligomers. Agents have been identified that interfere with, or inhibit, the oligomerization of the major component of amyloid plaques; a 42 amino acid long A&bgr;42 peptide product of proteolytic processing of the APP protein.
