1287796 6 - Salicylideneaminopenicillanic acid-salicylaldehyde adduct SANKYO CO Ltd 4 Nov 1969 [6 Nov 1968] 54056/69 Heading C2A Novel 6 - salicylideneaminopenicillanic acidsalicylaldehyde adduct is prepared by treating a 6-aminopenicillanic acid-containing fermentation broth at a pH between 6 and 9 with at least two moles of salicylaldehyde per mole of 6- aminopenicillanic acid. The adduct is isolated, or separated as a solution in a water-immiscible organic solvent for subsequent reaction with an acylating agent to produce a 6-acylaminopenicillanic acid. The fermentation broth may be a hydrolysed broth of a natural penicillin, e.g. penicillin G, and the reaction with salicylaldehyde may be carried out at 0-30‹ C. After the reaction the pH is brought to about 2 to 4 and a suitable water-immiscible solvent is added to extract the adduct. The organic phase may be washed, dried and evaporated to give the isolated adduct or may be treated with an acylating agent, e.g. an acid halide, active ester or anhydride or the acid itself in the presence of a condensing agent, to give the corresponding 6-acylaminopenicillanic acid. The freed salicylaldehyde may be recovered for further use.
