<p>PROBLEM TO BE SOLVED: To provide a culture method of Langerhans' islet capable of obtaining a sufficient amount of a transplant and an autotransplant. SOLUTION: Langerhans' islet is cultured in a culture blood serum (rat/ human) medium in which a radical scavenger, a growth factor, a matrix material, a nerve growth factor, a cell migration/scattering factor and anti-integrinβ-1 antibody are added in a culture process. At first, the medium is added with the radical scavenger and the growth factor, then cultured for about 12-24 h, and next, the matrix material, the radical scavenger, the nerve growth factor and a growth factor are supplemented. At 4-5 days during the culture process, the growth factor, the cell migration/scattering factor and the anti-integrinβ-1 antibody are supplemented. The culture process is continued for a long period of time so as every latent erythrocytes to be eliminated from the Langerhans' islet. The growth is continued for a long period of time for producing the islet in an amount sufficient to transplant for regenerating the islet for completely curing the patients with diabetes mellitus until showing normal blood glucose while not containing fibroblast.</p> |
