An apparatus/method (100) for securing large biochemical arrays has wide application in e.g. the fields of DNA fingerprinting and medical diagnostics. A light-source (106) irradiates the area under examination as part of a pixel array in which the target substance is caused to generate a fluorescent pattern for identification by comparison with standards. The incident angle of the light-flux (114) is such as to produce total internal reflection at the surface (112) of the transparent substrate (102) carrying the biochemical array (110) and is adjusted to give maximum pixel contrast of the fluorescent signals (116) for optical scanning (120) and comprises analysis (122).
