<p>PURPOSE:To provide the subject monoclonal antibody specifically reactive with the VLA-5 molecules, those present on the surface of mouse cells, applicable to analyzing mechanisms of the cell activation in immune system, wound curing, or metastasis, leading to development of medicines. CONSTITUTION:A rodent is immunized with the VLA molecule purified from the extract from mouse melanoma cells (B-16), the spleen is then taken out from the rodent, and a suspension of the splenocytes is made and mixed with mouse myeloma cells in the presence of a fusion promoter to effect cell fusion, and the resulting syncytia are diluted in a medium non-supportive for unfused myeloma cells and cultured. Thence, for the supernatant in each well containing the hybridoma, the presence of an antibody is confirmed using the reactivity with mouse melanoma cells as a marker to screen antibody-forming hybridoma followed by monocloning with limiting dilution technique, and antibody is then recovered from the culture supernatant for the resulting hybridoma, thus obtaining the objective monoclonal antibody specific to the mouse VLA-5 molecule.</p> |
