PURPOSE:To provide a plaster containing a keratolytic enzyme in a drug- containing layer or a part closer to the skin than the drug-containing layer. CONSTITUTION:A keratolytic enzyme (e.g. protease such as trypsin, pepsin, papain, esterase, bacterium-originated protease, etc.) is added to a drug- containing layer of a plaster or to a part closer to the skin than the drug- containing layer. The amount of the enzyme is 1/1,000-50IU(mg), preferably 1/100-10IU(mg) per 1cm<2>. The plaster produced by the above process is effective to promote the absorption of the drug into the body and to develop the pharma ceutical effect. It is preferable to add a part of the substrate and a coenzyme, etc., to the enzyme to improve the activity. Preferably, the plaster is added with a pH-modifier to improve the enzymatic activity. EFFECT:Enzyme is hardly absorbable in the body and the drug action can be controlled freely to a desired state such as rapid-acting action, durable action, etc.
