PURPOSE:To determine the base sequence of a nucleic acid with high accuracy by obtaining an electrophoretic image reduced in distortion, by preparing the plate shaped body for covering an electrophoretic support from a good heat conductive material and attaining to uniformize the temp. distribution of the electrophoretic support. CONSTITUTION:An electrophoretic support (comprising acrylamide or the like) 2 is held between two electric insulating plates (glass plates) 3. Plate shaped bodies 5 comprising a good heat conductive material (copper, aluminum or SiC) is provided on said insulating plates 3 so as to cover both of them and thermostatic apparatuses 4 are further provided on said plate shaped bodies. Electrophoretic buffer solution tanks 1 are provided to both ends of the electrophoretic supports 2 and an electric field is formed by a high voltage DC power source 6 to form an electric field. Then, a nucleic acid specimen is injected in the electrophoretic support 2 to be migrated. Because the temp. distribution of the electrophoretic support 2 is uniformly controlled by the thermostatic apparatus 4 and the plate shaped bodies, the specimen migrates in such a state that the distortion of the electrophoretic separation zone is reduced. Therefore, the determination of the base sequence of the nulceic acid can be made highly accurate and automatic. |
