摘要 PURPOSE:To carry out determination of N-acetylneuraminic acid in high accuracy by a simple operation without being influenced by endogenous pyruvic acid, by treating a specimen of N-acetylneuraminic acid with an enzyme such as neuraminate aldolase, acylglucosamine 2-epimerase, etc. CONSTITUTION:N-Acetylneuraminic acid is decomposed with neuraminate aldolase into N-acetylmannosamine and pyruvic acid. Prepared N-acetylmannosamine is treated with AG-epimerase, etc., to prepare N-acetylglucosamine. Then, this N-acetylglucosamine is treated with N-acetylhexosamine oxidase in the presence of O2 and water, to give N-acetylglucosaminic acid and H2O2. Any of N2O2, N- acetylglucosaminic acid and consumed O2 is measured, to determine N-acetylneuraminic acid.
