PROBLEM TO BE SOLVED: To provide a sealing structure of an opening and closing cover which is excellent in follow-up performance according to elastic deformation to heighten sealing performance and air tightness. SOLUTION: This sealing structure of the opening and closing cover is provided with an opening and closing cover 1 for openably and closably blocking up an opening part of a contained to be sealed, an annular groove 6 having a substantially inverted-trapezoid-like cross section, which is formed on the peripheral edge part 1a of the opening and closing cover opposite to the opening part and having an inclined inner surface 6c inclined inward to be fan-shaped, and an annular elastic seal member 11 removably fitted to the annular groove. The elastic seal member is provided with an annular base part 11a having a substantially rectangular cross section removably fitted from an opening 6a of the annular groove and having the end face abutting on the bottom 6b of the annular groove, an expanded part 11e projected on the annular base part and abutting on the inclined inner surface of the annular groove, and a wedge-like tip part 11e exposed from the opening of the annular groove, projected on the expanded part side of the annular base part and having the tip part 11d formed substantially flat. When the opening and closing cover is blocked up with the wedge-like tip part pressed to the opening part of the container to.be sealed, the annular base part of the elastic seal member is elastically deformed to press the wedge-like tip part to the opening part of the container to be sealed, thereby accomplishing sealing.
