<p>1,273,025. A 17α- (1 - methy(propadienyl) steroid. SANDOZ Ltd. 2 April, 1969 [10 April, 1968; 12 Sept., 1968], No. 49412/71. Divided out of 1,273,024. Heading C2U. The novel compound 17α-(1-methylpropadienyl) - 17# - hydroxyestr - 4 - en - 3 - one is prepared from estr-4-ene-3,17-dione by reaction with an appropriate organometallic reagent to introduce the 17α-(1.methylpropadienyl) or a 17α-(1-methyl-2-propynyl) group, the latter group then being subjected to basic isomerization. The 3-oxo-#<SP>4</SP> grouping is protected during the organometallic reaction, e.g. as 3-alkoxy- #<SP>2</SP>'<SP>5</SP>(<SP>10</SP>) or 3,3-ethylenedioxy-#<SP>5(10)</SP>. The 17#- hydroxy group is protected during base isomerization, e.g. as methoxy or tetrahydropyranyloxy. When final removal of a protecting group results in the #<SP>5(10)</SP>-isomer of the desired product, acid isomerization is effected accord ingly. The novel compound is said to possess progestational activity and may be made up with carriers into pharmaceutical compositions for oral and parenteral administration.</p> |
