摘要 <p>Assaying a plasma NADH-dependent catalytic enzyme (EN) in a sample of biological fluid by measuring its enzymatic activity, comprises measuring the reduction of NADH fluorescence, where the biological fluid is total blood and the enzymatic activity of the catalytic enzyme is measured by monitoring the reduction of the NADH fluorescence in a microfluidic chamber having a thickness of 20 mu m to 2 mm. Independent claims are included for: (1) process-I for assaying, in a biological fluid sample, a plasma catalytic enzyme (Ec) having as substrates S1 and S2 by measuring the enzyme activity of the enzyme (Ec), comprising: (a) incubating the biological fluid with the substrate S1, NADH, one or more different NADH-dependent catalytic enzymes having substrate for one of the enzymatic reaction products catalyzed by Ec, and optionally pyridoxal phosphate (PLP); (b) adding the substrate S2 for initiating the enzymatic reactions catalyzed by catalytic enzymes Ec and EN, where the biological fluid is total blood, in that after adding the substrate S2 in step (b), the resulting mixture is immediately introduced into the microfluidic chamber and the enzyme activity of the catalytic enzyme Ec is measured by following the reduction of the NADH fluorescence in the microfluidic chamber; (2) kit or pack for assaying a plasma catalytic enzyme (Ec) in a total blood sample, the enzyme having as substrates S1 and S2 , comprising a microfluidic chamber, and following reagents, the substrates S1 and S2, the NADH, at least one NADH-dependent catalytic enzyme, and optionally PLP; and (3) kit or pack for assaying a plasma catalytic NADH dependent (EN) enzyme in a total blood sample, comprising a microfluidic chamber.</p>
