PURPOSE: An analytical method of lyso-globotriaosylsphingosine in urine by an LC-electrospray Ionization tandem mass spectrometry and a diagnostic method of fabry disease using the same are provided to conduct a pretreatment process by a lower scale. CONSTITUTION: An analytical method of lyso-globotriaosylsphingosine in urine uses a liquid chromatography-electrospray Ionization tandem mass spectrometry (HPLC ESI MS/MS). The conditions of the liquid chromatography are followings. Mobile phase composition : mobile phase A (50% or less of acetonitrile with added formic acid), mobile phase B (acetonitrile of 100% with added formic acid). Mobile phase concentration gradient: start - 0.1 minutes : maintaining 20% of mobile phase B, 0.1 minutes - 1 minute: increasing the mobile phase B from 20% to 90%, 1 minute - 2 minutes : maintaining 90% of the mobile phase, 2 minute - 5 minutes : decreasing the mobile phase B from 90% to 20%. Mobile phase velocity : 200 ML - 1 mL/minute. |
