A method for separating bio-substances for mass spectrometric analysis comprises at least first and second fractionations, each fractionation comprising applying a solution containing bio-substances to a binding surface, removing the solution and any unbound bio-substances from the binding surface, collecting said unbound bio-substances, applying a solvent composition to the bound bio-substances and collecting them and ten applying different solvents as required so as to remove target bio-substances from the binding surface. The bio-substances may be a mixture of proteins and/or protein conjugates. The binding surface may be a selective binding surface, an affinity coating or a solid phase usable in liquid chromatography. The method may be carried out by a pipetting robot. A microtitration plate 1 suitable for the method comprises microvessels 2 having binding surfaces which are sealed with a piercable foil. A kit for carrying out the method comprises a microtitration plate and at least one pipette tip 5 for piercing the foil, which may be profiled so as to correspond to the shape of a microvessel on the microtitration plate. |
