PURPOSE:To stably measure unsaturated iron bonding ability in blood, by reducing trivalent iron in the blood with an oxidase into bivalent iron in an enzymic reaction system and colorimetrically determining the formed color of a complex of the bivalent iron with a chelate color former. CONSTITUTION:An enzyme and a substrate therefor are separately added to prepare the first or second reagent. Trivalent iron and color former are separately added to prepare the first or second reagent. The reagent containing the trivalent iron is reacted with blood serum and the reagent containing the color former is added and reacted therewith. As a result, unsaturated transferrin in the blood serum is bound to the added iron to provide saturated transferrin and unbound iron is reduced from the trivalent iron into bivalent iron and bound to the color former to assume a color. The amount of the unbound residual iron is subtracted from the known excess amount of the iron to calculate the unsaturated iron binding ability.
