PREPARATION OF ERYTHROPOIETIN-PRODUCING CELL

摘要

PURPOSE:To prepare an erythropoietin-producing cell, by introducing a mixed vector of an erythropoietin transducing vector and selected marker gene transduced vector into a mouse L929 cell using a specific technique. CONSTITUTION:An aminoglycoside 3'-phosphotransferase gene (neo gene) derived from Tn which is a selected marker is inserted into the downstream of SV40 early gene of a shuttle vector pKSV-10 to prepare a selected marker neo gene transduced vector pKSVNeo, which is then mixed with an erythropoietin transducing vector pSVhEPX. The resultant mixed vector is further introduced into a mouse L929 cell by a DNA transfection technique using calcium phosphate to prepare the aimed erythropoietin-producing cell.
