PURPOSE:To exactly measure endotoxin with an extremely simple operation by measuring the ammonia which is generated by bringing a specimen into reaction with the blood cell extract of Limuls polyphemus (horseshoe crabs) and specific substrate by an electrode means for detecting ammonia. CONSTITUTION:A 10mM phosphoric acid buffer soln. contg. 0.6% NaCl is added at 2ml into an ampoule of Reissert to dissolve the freezedried Reissert and 416mug (208mug/ml) substrate expressed by the formula (I) is added thereto; further the endotoxin is added thereto to make 0, 10, 10<2>, 10<3>, 10<4>, or 10<5> pg per ml. The reaction is caused for 90min at 37.5 deg.C. An ammonia electrode is inserted therein and the potential is read after 10min of equilibration. As a result, a direct association is admitted between the logarithm of the concn. of the endotoxin and the potential of the ammonia electrode in the 10-10<3>pg/ml range of the endotoxin concn.
