<p>The process consists of (a) purifying the lipid material contaminant from a plasma pool from chronic carriers of AgHBs by treatment with Freon 113 in amt. 4% for 30 minutes at 20 deg.C; (b) reabsorbing the AgHBs on Aerosil in concn. of 2-10 w/v % and incubating for 10-25 hours at 0-40 deg.C; (c) eliminating residual proteins by centrifuging for 15 mins at 1500 g; (d) washing the contaminant proteins from the Aerosil with a low-strength ionic buffer, e.g. saline phosphate (140 mM NaCl and 10mM sodium phosphate, with pH 7.4); (e) eluting the AgHBs from the adsorbent with borate buffer (0.015 moles and pH 9.3) for 1 hour at 37 deg.C and centrifuging; (f) neutralising the supernatant liquid with HCl and purifying with polyethylene glycol of mol. wt. 6000 to give a final concn. of 9%; and (g) ultra-centrifuging and repeating the process.</p> |
