Antibiotic NK-1001 is produced by cultivating a kanamycin-producing strain of Streptomyces kanamyceticus under aerobic conditions in a culture medium containing assimilable sources of carbon and nitrogen in the presence of streptidine; 0À05 to 1À0% and preferably 0À1 to 0À5% by weight of streptidine is added to the culture medium, preferably after 80 to 90 hours cultivation, the duration of cultivation being 4 to 8 days. Alternatively, an antibiotic NK-1001-producing mutant of Streptomyces kanamyceticus ATCC 21252, namely mutant ATCC 21257, may be cultivated in conventional medium, i.e. in the absence of streptidine. Antibiotic NK-1001 may be isolated from the clarified broth by adsorption on "Amberlite" IRC-50, followed by elution with 0À5N aqueous ammonia, concentration, dissolution in water, treatment with active carbon, filtration, treatment with "Amberlite" CG-50 and elution with 0À3N aqueous ammonia. After freeze-drying, the power was dissolved in water and methanol added to produce crystals of NK-1001. Antibiotic NK-1001 is a basic, broad spectrum antibiotic related to the kanamycins in that it consists of 1 mol. deoxystreptamine, 1 mol. 6-amino-6- deoxy-D-glucose and 1 mol D-glucose; empirical formula C 18 H 35 N 3 O 12 .H 2 O; shows no U.V. spectrum; infra-red spectrum as in Fig. 2; [α]21 D = + 132 degrees (1% aqueous solution); colourless needles m.pt. 238-242‹ C.; soluble in water, aqueous methanol, aqueous ethanol; insoluble in anhydrous methanol or ethanol, ethyl acetate, butyl acetate and benzene. Antibiotic NK-1001 may be extracted from the culture broth with an organic solvent containing a carrier such as p-toluene sulphonic acid or lauric acid.
