<p>The present invention relates to a method for detecting herpes simplex virus type-1 and type-2 in a biological sample, characterized by the steps of extracting deoxyribonucleic acid; amplifying a region of the deoxyribonucleic acid by contacting the deoxyribonucleic acid with a first pair or a second pair of amplication primers, or a combination of the first pair and the second pair of amplification primers in the presence of a fluorescent dye; wherein the first pair of amplification primers comprises a forward primer (SEQ ID NO: 1) and a reverse primer (SEQ ID NO: 2); and the second pair of amplification primers comprises a forward primer (SEQ ID NO: 3) and a reverse primer (SEQ ID NO: 4); detecting the fluorescence generated during amplification; subjecting the amplified deoxyribonucleic acid to gradually increasing temperature and simultaneously monitoring the fluorescence; determining the presence of herpes simplex virus type-1 and type-2 in the biological sample by detecting a sharp reduction of fluorescence when the temperature reaches 86-88°C for herpes simplex virus type-1 and 88-89°C for herpes simplex virus type-2 during gradual temperature increase after amplification.</p> |
