PROBLEM TO BE SOLVED: To provide a preparation process of a cellulose microporous film which improves the flatness while increasing the yielding rate of nucleic acids. SOLUTION: The cellulose microporous film, a saponified film 12, is prepared by performing each process of saponification, neutralization, washing and drying while transporting a microporous film of a cellulose ester derivative. Water to be used in the process is free from nuclease. The drying device 18 is equipped with a drum 25 and a belt 26, and the saponified film 12 is sandwitched between them to be transported and to be heated. The transporting rate is 0.05-5.0 m/minute. The drying process is performed in a clean room with the cleanliness class of at least 7 measured by ISO 14644. The saponified film 12 prepared by the above has an excellent flatness and keeps the flatness when it is cut into a circle with the diameter of 7 mm.
