PURPOSE:To obtain a looped DNA mixture produced by partially converting a single-stranded DNA of a bean golden mosaic virus into double-stranded structure, having a specific gene map and useful as a vector for a gene manipulation of plant. CONSTITUTION:This mixture containing the monomer to trimer of DNA having a gene map of formula I (P1 is 5'-AAGCTT-3', P2 is 5'-ATCGAT-3'; P3 is AGATCT-3'; P4 is 5'-GTCGAC-3'; P5 is 5'-CPyCGPnG-3'; P6 is 5'-ATCGAT-3'; P7 is 6'-GTCGAC-3') and the monomer to trimer of DNA having a gene map of formula II (P8 is P1; P9 is P2; P10 is P4; P11 is P4; P12 is P1; P13 is P3) is produced by purifying bean golden mosaic virus (BGMV) from a plant infected with the virus, separating a single-stranded DNA from the virus by conventional method and reacting the DNA in the presence of a DNA polymerase using a calf thymus oligonucleotide as a primer to convert the single-strand into a double-strand and obtain the objective DNA having the single-stranded DNA of BGMV partially converted to double-stranded DNA. |
