MONOCLONAL ANTIBODY AGAINST HUMAN IL-2 RECEPTOR BETA CHAIN

摘要

PURPOSE:To obtain a monoclonal antibody by fusing an antibody-forming cell of mammals except human immunized against beta chain manifesting cell strain YTS with a cell having permanently multiplying properties to give a hybridoma and culturing the hybridoma. CONSTITUTION:An antibody-forming cell of mammals except human immunized against beta chain manifesting cell strain YTS is blended with a cell having permanently multiplying properties and subjected to cell fusion by gradually adding a fusion promoter such as polyethylene glycol while maintaining the reaction mixture at about 37 deg.C. Then the fusion promoter added to the culture solution is diluted to stop the fusion and the cell is separated by centrifugal separation. The cell is cultured in a hypoxanthine aminopterin thymidine to give a hybridoma. The prepared hybridoma is cultured in vitro or in vivo to produce a monoclonal antibody.
