PURPOSE:To make it possible to determine a substance having colony forming ability in a short time and automatically, by measuring absorbance of a formazan compound produced by participation of mitochondria in a mouse myeloid cell. CONSTITUTION:In a method of measuring colony stimmulating factor by treating a mouse myeloid cell with a colony stimmulating factor and multiplying the cell, first, a tetrazolium compound is converted into a formazan compound by participartion of mitochondria in the cell. Then absorbance of the formed formazan compound is measured to determine activity of the colony stimmulating factor. The above-mentioned measuring method can be usually carried out as follows. (a) The tetrazolium compound is added to the multiplied mouse myeloid cells and (b) the tetrazolium compound is converted into the formazan compound in the cells by participation of mitochondria. (c) The formazan compound is eluted in the cells, homogenized and (d) absorbance of the eluted formazan compound is measured.
