PROBLEM TO BE SOLVED: To obtain a novel IL-6 receptor-IL-6 direct-binding fused protein which is obtained by directly binding one of amino acid residues constituting IL-6 receptor with one of amino acid residues constituting IL-6, possesses low antigenicity and is useful as a hemopoitic stem cell-amplifying agent, a platelet- increasing agent or the like. SOLUTION: This fused protein is a novel IL-6 receptor-IL-6 fused protein obtained by directly combining one of amino acid residues constituting interleukin-6(IL-6) receptor with one of amino acid residues constituting IL-6, allows to expect high therapeutic activity and decreased antigenicity in comparison with an IL-6 receptor-IL-6 fused protein containing a linker sequence and is useful as a hemopoietic stem cell-amplifying agent ex vivo and a platelet- increasing agent. This fused protein is obtained by successively integrating a gene encoding IL-6 receptor and a gene encoding IL-6 into a vector, transducing the vector into a yeast belonging to pichia pasturis species to transform followed by cultivating this yeast and separating an expressed product from the cultured cell. |
