PROBLEM TO BE SOLVED: To obtain a new sciatic nerve ciliary neurotrophic factor having each specific amino acid sequence, molecular weight and specific activity, with its biological activity resistant to reducing agents and SDS, and useful as e.g. a therapeutic agent for neurodegenerative diseases such as Alzheimer's syndrome. SOLUTION: This factor, which is a substantially purified new proteinic sciatic nerve ciliary neurotrophic factor (SN-CNTF) such as a human sciatic nerve ciliary neurotrophic factor having a molecular weight of about 24,000 determined by SDS-polyacrylamide gel electrophoresis, and a specific activity of 2×10<8> TU/mg or higher, with its biological activity resistant to SDS and reducing agents, and having an amino acid sequence of the formula, is useful as e.g. a therapeutic agent for neurodegenerative diseases such as Alzheimer's syndrome, Parkinson's disease and multiple sclerosis each developed due to the death of neurocytes or gliocytes. The factor is obtained by integrating an expression vector with a gene coding for SN-CNTF afforded by screening an animal or human genome library followed by expressing the gene in host cells. |
