<p>It is intended to embody a method of conveniently detecting 5-methylcytosine within a short period of time at a low cost and a high sensitivity and a detection kit. A conventional invader method, whereby the presence or absence of the methylation of a base cannot be detected, is modified and combined with a method of selectively cleaving a 5-methylcytosine site in a target DNA. More specifically speaking, the 5-methylcytosine site in the target DNA is selectively cleaved to give a 5'-fragment. Then, an enzymatic reaction is conducted by using an FRET probe hybridizable with this 5'-fragment and flap endonuclease and the fluorescence is detected.</p> |
